Test Method Validation (TMV) demonstrates, with evidence, that a laboratory method is fit for its intended use. Whether the method is chromatographic, spectroscopic, microbiological, physical (e.g., torque, hardness), or vision/algorithm‑based, TMV proves the method can reliably distinguish conforming from non‑conforming product within defined ranges, matrices, and operating conditions. TMV sits at the heart of Quality Control (QC) and informs release decisions alongside routine laboratory analyses & review, trending, and stability knowledge.

1) Define intended use & risk. Clarify what the method decides (release, IPC, stability), target specification, and risk in the Risk Register.
2) Develop & pre‑validate. Optimize conditions; run pilot studies to tune accuracy, precision, and specificity; draft acceptance criteria grounded in science.
3) Execute TMV. Perform the full protocol across analysts/instruments/days; confirm system suitability protects daily operation.
4) Control & maintain. Release the method under SOP; trend performance in LIMS; manage changes via Change Control and periodic review.

If a criterion fails, open a Deviation, investigate root cause, apply CAPA, and repeat with traceable data segregation.

5) Method Characteristics—Getting the Science Right

Accuracy is demonstrated with traceable standards or spiked matrices; precision is split into repeatability (same day/analyst) and intermediate precision (days/analysts/instruments). Specificity (or selectivity) proves the method distinguishes analyte from interferences, including degradation products and matrix effects. Linearity and range confirm proportional response where required; LOD/LOQ define detection and quantitation boundaries with appropriate statistical treatment. Robustness trials deliberately vary small factors (e.g., column lot, flow, temperature) to show the method stays on‑spec. Methods evolve. Changes to reagents, columns, software, algorithms, ranges, or matrices must pass through Change Control with risk assessment defining whether revalidation is required. Link method versions to batch records and reports so results remain interpretable over time. Periodically review performance and feed lessons into the Process Control Plan and APR.

12) Method Transfer, Outsourcing & Supplier Control

When methods move between sites or to contract labs, perform method transfer/verification with predefined acceptance criteria. Align responsibilities in the Quality Agreement and manage labs under Supplier Quality Management (SQM) and Supplier Qualification. Differences in instruments or software should be bounded by robustness data and captured in site‑specific system‑suitability criteria.

13) Metrics That Demonstrate Method Control

  • Weak integration rules. For chromatographic/vision methods, lock integration/model rules and review changes under Document Control.
